Pick your strongest team and tactics that suit, taking into account your opponents strengths. Also have a look at the list of available players, or if you haven't got much money check out the bank tab & see if it's worth getting a loan to buy new players.

Without sharing your user publically, I'm taking a guess at which user you are Rob. If I'm right, you have joined the TWTD League, and when logged in, click on "Play". You can then see your squad, etc, etc.
(You might have clubs in various leagues, hence the need to click Play button)

One change made at the end of last season was to the transfer list - to make it fairer, and I personally think more fun... - you now bid for players and the winning bid gets the player after the next match. Check it out :)
